Someone sent me a v5 file yesterday. I converted the Y-DNA SNPs that have a call then I pulled just the SNPs that are P310 and below. I set the filter to just show SNPs that have include the DF27 ISOGG 2017 longhand name of R1b1a1a2a1a2a.

These are the DF27 SNPs with a call included in the 23andme v5 raw data.

I attached the spreadsheet that I had created so that the other included information can be seen. I am looking forward to seeing people positive for the above SNPs showing up at 23andme in the future. Hopefully 23andme allows for an upgrade in the future. That way people that don't want to, or can't, test elsewhere are able to see if they are positive for any of them.


P312 and Z195 aren't tested but the following are so many of the people that show "terminal" for P310 or P311 at 23andme are even more likely to be positive for DF27 than people that currently show 'terminal" for R-L51.

I had some time today to pull data from Alex Williamson's BigTree to get an idea of what percentage of DF27 people will get a DF27 subclade through 23andme v5 and which percentage won't. There are currently 767 people in the BigTree that are positive for DF27. There are 329 people that are positive for the DF27 SNPs tested by 23andme which is almost 43%. So likely there will be more than about 57% of the testers at 23andme that should be in a subclade of DF27 that will be given a "terminal" SNP of P310 or P311.

23andme tells me R-P311 but I've got the two SRY2627/M167 SNPs it tests for.

Do you have G or A for i3000043 and i701489? You should have G.

Both i3000043 and i701489 are actually for the same SNP since it is the same position of 2658271.

Yes it does and so does the Ybrowse site at http://ybrowse.org/gb2/gbrowse/chrY/?

There are people with v4 testing that show positive for SRY2627 at 23andme so I wonder why 23andme isn't showing you positive for that SNP. I hope that they fix the problem soon. Have you contacted 23andme about the problem?

23andme's non-responsive response:

You may be able to find additional information through resources outside of 23andMe, though we cannot validate the accuracy of that information.

I don't see the old concatenated notation at all at 23&me. Many, I think most, people who are positive for df27 will be indicated as L51 at 23&me.
A bit less will be indicated as P311, and still much less as P312.
I know 2 cases of people tested df27 at FTDNA who are just given L51 at 23&me : an autosomal match of mine who is by chance part of the df27 group at FTDNA, and myself.

But I am curious about these V5 people, does 23&me show their terminal called SNP, or will it show only a much higher level, like P312 or even DF27, even when positive for one of the SNP indicated above ?

They got rid of it with the New Experience.


Many, I think most, people who are positive for df27 will be indicated as L51 at 23&me.
Not with the v5 chip they won't. They will be shown to be R-P311 like djp5 until they update the report.


A bit less will be indicated as P311, and still much less as P312.
All DF27 people on the v5 chip will be shown as R-P311. R-P312 isn't even tested by v5.


I know 2 cases of people tested df27 at FTDNA who are just given L51 at 23&me :
Most DF27 people that show to be L51 were tested by v4. The v3 test included L151 which is a phylogenetic equivalent to P311. The v2 test included P312. However, this thread is about v5.

They will be shown to be R-P311 like djp5 until they update the report. They do not show their terminal called SNP. 23andme has updated quite a few things about Y-DNA but they don't include a lot of SNPs downstream from R-P311 in the report for v5 tested people.

Back in the day, Adriano Squecco maintained a spreadsheet of people who had sent him their 23andMe raw data (from Illumina chips, I guess v3 and v4). With some cooperation from David Reynolds and a few others he was finding a good many SNPs pointing to known subclades, including DF27 subclades. That was where my own L484+ turned up. L484 itself proved too recurrent and impermanent to be a reliable subclade marker, but some good SNPs were also identifiable.

Recently someone told me he had one of the subclades of the Rox2 Cluster below DF27 identified (by a geneticist friend) in his 23andMe results. He's getting a BigY at FTDNA, to see if it's real; neither I, nor (I believe) he, can confirm it yet. But it's another approach. There is no reason to assume that the 23andMe folks know, let alone report, everything that's being discovered by their own test. Although with some chips, they are only going to find what the chip has been told to look for. I don't know anything really current about 23andMe, though I believe they are no longer using Illumina technology. That probably makes a difference, but I'm not sure whether it's better or worse.

I am more interested in the known SNPs that are readily available at both ISOGG and Ybrowse as opposed to what is discovered since very few SNPs will be discovered by 23andme anyway. 23andme has been updating the Y-chromosome page info by using a "terminal" SNP and providing a tree in the scientific details of the paternal haplogroup report at https://you.23andme.com/reports/paternal_haplogroup/ Since they have been updating the Y-chromosome info they should have a tree that has all of the SNPs that are available at ISOGG. Even if they used the 2016 tree I would be happy with that.

These are the DF27 SNPs, that I found, with a call included in the 23andme v5 raw data by using the Ybrowse data that has a longhand ISOGG haplogroup as a reference. Since ISOGG has all of them, or almost all of them in case I missed one, then 23andme should be reporting these SNPs, for v5 customers, as a paternal haplogroup, or at least as a subclade, when the customer is positive for one but they don't.

Since you mentioned another SNP I pulled the dataset from Alex Williamson's BigTree and used it as a reference and I found some more. So if I use his data it is more reliable since there are less back mutations that confuse the results. However, I had to do a manual search of the SNPs to determine which additional SNPs are at ISOGG.

I don't know anything really current about 23andMe, though I believe they are no longer using Illumina technology. That probably makes a difference, but I'm not sure whether it's better or worse.

"Know about" is a little strong, I have a rumor in email from an FTDNA DF27 project member who is invisible until the transferred kit has some STRs tested there. A friend looked at his (or her) brother's 23andMe raw data:

"The SNP this amateur friend found in my brother's raw data is this:

Rs2704748 y 22288829G

Whatever it is and what it means, I don't know. Obviously, he could be wrong and it may not even be a DF27 at all."

I don't know either, a G at that position doesn't look like a SNP to me (for a P312 guy, anyway). I referred this member to Chris Corner for Rox2 verification, or not.


23andme v5 uses the Illumina GSA chip which is the same one used for LivingDNA. https://cruwys.blogspot.com/2017/08/23andme-launch-new-v5-chip-and-revise.html It has a lot more Y-DNA SNPs than chips previously used by 23andme.

OK, I was just misinterpreting stuff I had seen in the chatty forum 23andMe provides (but doesn't moderate, I think). Illumina v3 and v4 non-medical raw data can be synched with FTDNA Family Finder, but data from their newish v5 can't be. I inferred (incorrectly) that they had changed chip makers.

Finally, I was pleased to see that FGC15710 is detectable with the new chip. (My test there was on v3, and they have me classified as R-P311.) The FGC157xx series of SNPs below Z220 were discovered in my own sample at FGC. The branches below FGC15710 have never been submitted to ISOGG, but they are on Alex's Big Tree and the YFull tree. I've also been tested at LivingDNA -- if they are using that same chip, they are unaware of anything below Z220 it finds, or should find, in my results (or, presumably, anyone else's). That would include the Z295 and CTS4065 levels, not just FGC15710 and below.

PS -- Armando, I think you should post your revised list on the 23andMe forum also -- either as a new thread, or an update to the one started Sept. 5 by "Julius" that is titled
Paternal haplogroup is R-P311- can barely find anything about it? I think it deserves its own thread. The members of that forum who ought to see it may, however, never have heard of DF27 -- so perhaps the title really should include R-P311, or maybe both.

"Know about" is a little strong, I have a rumor in email from an FTDNA DF27 project member who is invisible until the transferred kit has some STRs tested there. A friend looked at his (or her) brother's 23andMe raw data:

"The SNP this amateur friend found in my brother's raw data is this:

Rs2704748 y 22288829G

Whatever it is and what it means, I don't know. Obviously, he could be wrong and it may not even be a DF27 at all."

I don't know either, a G at that position doesn't look like a SNP to me (for a P312 guy, anyway). I referred this member to Chris Corner for Rox2 verification, or not.
RS2704748 22288829 G is FGC12056 and is found in v4 data but not in v5 data and it is a false positive in v4 since I also have G for RS2704748 at 22288829.



Finally, I was pleased to see that FGC15710 is detectable with the new chip. (My test there was on v3, and they have me classified as R-P311.) The FGC157xx series of SNPs below Z220 were discovered in my own sample at FGC. The branches below FGC15710 have never been submitted to ISOGG, but they are on Alex's Big Tree and the YFull tree. I've also been tested at LivingDNA -- if they are using that same chip, they are unaware of anything below Z220 it finds, or should find, in my results (or, presumably, anyone else's). That would include the Z295 and CTS4065 levels, not just FGC15710 and below.
Even though 23andme v5 and LivingDNA use the same chip the chips can be customized for the customer and therefore some SNPs can be left. However, LivingDNA has had problems reporting some positive and negative SNPs for some customers so I recommend sending them an email to see if you the test included a result for Y-chromosome position 23067644 hg19 and if so what the result was and to please reassess the results of other customers since it is FGC15710 in hg19. I mentioned hg19 since the Ybrowse website is now using hg38 but they still provide the hg19 file.

I did look for SNPs downstream from FGC15710 using Alex's SNPindex in the 23andme v5 raw data but I didn't find any.


PS -- Armando, I think you should post your revised list on the 23andMe forum also -- either as a new thread, or an update to the one started Sept. 5 by "Julius" that is titled I think it deserves its own thread. The members of that forum who ought to see it may, however, never have heard of DF27 -- so perhaps the title really should include R-P311, or maybe both.

I have been planning to update the one that I point people to which is a different one. I need to get organized first. I'm hoping that people realize that they need further testing even if there is a DF27 SNP tested by 23andme v5 or not.

My Living DNA results I just received. My fatherline came back as P312.
I have to assume the Illumina GSA chip does not test for DF27 or Z2573.

No it does not test DF27 nor does BigY the vast majority of the time. There is a special test for DF27 that has to be ordered individually or is tested by some SNP packs testing negative for certain other SNPs. When tested positive for SNPs downstream from DF27 there is no need to test for DF27.
