So according to 23andme V4 chip my late maternal grandfather's sister is U4b, and so he was, but I ran her mt-DNA through Jameslick software, and this is what I got:

Markers found (shown as differences to rCRS):

HVR2: 73G 215G 263G 499A
CR: 750G 1438G 1811G 2706G 4418C 4646C 4769G 5999C 6047G 7028T 7403G 7705C 8860G 11467G 11696A 11719A 12308G 12372A 14620T 14766T 15693C
HVR1: 16129A 16356G (16519C)

IMPORTANT NOTE: The above marker list is almost certainly incomplete due to limitations of genotyping technology and is not comparable to mtDNA sequencing results. It should not be used with services or tools that expect sequencing results, such as mitosearch.

Any help as to where this is common would be more than welcomed?

So I was looking at 23andme and it seems my great aunt is ancestral for the U4b1 and U4b2 markers, 23andme doesn't test for U4b3, she is definitely within the U4b cluster though, and within the U4 cluster(5+ mutations confirmed). She has 500+ cousins yet no one has U4b as mt-DNA. So I am once more wondering from where her U4b(xU4b1,U4b2) comes from?

So it seems the "Flips(1): 16356G" is artificial, in reality what is happening is that in James Lick program he has U4+ as being defined by HVR1: 16356C, but in reality the i3001881 (https://www.23andme.com/you/explorer/snp/?snp_name=i3001881) is defined as T16356G per 23andme and A16356C per Cambridge Reference Sequence, so it is in reality not really a flip. Now what I found interesting is the A263G or rs2853515 (https://www.23andme.com/you/explorer/snp/?snp_name=rs2853515), that is one of the defining markers of U4b3, so perhaps she is U4b3 since 23andme doesn't test for it. She has 5 extra mutations from U4b3, and 6 extra mutations from U4b, this is not a full mitochondrial genome, so I'm guessing both U4b and U4b3 are rather old clades. Does anyone know anything about U4b or U4b3?

What's the distribution of U4b3?

Greetings.

I am U4b3 through my maternal grandmother who was born in 1892 in what is now Valmiera, Latvia. She arrived in Australia in 1927 as a refugee and I have no details of her ancestry.

According to the following, U4b3 is found in Russia, Greece and Austria.

Hope this helps.
