Familytreedna announced a Big y sale, price will be reduced to 495 and a coupon earned during father day mat be used. Im still L2* and im looking for such a coupon to order the big y. Who can help me?

Unfortunately, I have not been able to find any more coupons. For anyone who needs help paying for the test and is U152+, we can contribute $50 from the project funds per Big-Y order. Please send me an email if you are interested.

Thanks, Richard. I just ordered BigY before I saw your kind offer. I am U152+, Z49+, S8183+ and awaiting some more downstream results from YSeq. I've posted elsewhere about our project to test the remains of Richard Barry, 6th Earl of Barrymore(https://sites.google.com/site/barrymorednaproject/) and am ordering BigY for myself in preparation for obtaining those ancestral results for comparison.

Thanks to some friends I also ordered the Big-Y and my batch is 586
I am very excited but the results are expected in 8-10 weeks.

My results came in today!

I'm not very good in evaluating my results but so far i notice i'm negative on all known downstream snp from L2. But dont know if something new was discovered

Wow! That seems like a really fast turn around time for the lab. Are a lot of results coming in this quick? I noticed Sven's last post in this thread was from Aug 31st saying he just ordered it.

Thank you, i was also suprised they came in since te expected date was only 31/10; that's 2 weeks earlier. I hope Rocca can help a bit to analyse the results.

My Big Y results are in. Kit number 240232. Looks like an exact SNP match to Petr, 195364. I've requested the .bam file. In the meantime, Richard I have sent Steve an email and I would appreciate any help you or he might give me with interpretation or guidance on further testing.

Your welcome! I thought that was quick! Your post prompted me to email my brother's closest 67 marker match who ordered the test during the June sale.

He just told me that his results came in a couple weeks ago. But he only sent his bam file to one of his project administrators a few days ago. So I am looking forward to his results. :)

Thanks, Richard. In my BigY matches I differ from Bruns on 2 SNPs, F2764 and Z2001. I have an exact SNP match as I mentioned with 195364, who has Czech ancestors. When I ran an STR comparison, the TMRCA ranged from 2200 years (95%) to 1600 years (50%) bp. That's interesting because my ancestors were Anglo-Norman-Irish, with a possible origin in Flanders. That brings up one potential scenario involving the Boii Celts, who were supplanted in their eponymous Bohemia by Germanic tribes. There probably were some who remained in what is now the Czech republic but others moved west and settled in what became Cisalpine Gaul. In the first century AD they participated in the Battles of Bibracte and Alesia, within a reasonable distance of Flanders and Normandy. No way to know for sure but interesting nonetheless.

I'll try to get some of the other Z49+ men in the Barry surname project to do a BigY. We may uncover some private family SNPs.

Kit 348783, delivered to the lab on June 25th, batched on July 10th as batch # 578, no results yet, to my yesterday question I've got a reply that there is some quality problem.
Kit 348788, delivered to the lab on June 25th, batched on July 3rd as batch # 577, results received yesterday after second question to the support.
Kit 348790, delivered to the lab on June 25th, batched on July 3rd as batch # 577, results received on September 30th, still resolving bogus BigY matches with the support.
Kit 348792, delivered to the lab on June 25th, batched on July 10th as batch # 578, results received on September 30th, still resolving bogus BigY matches with the support.

Hi Richard, what does this mean exactly? Does it mean that Jim's results for all my positive SNPs 17760314(T/C), S12993, 8356636(T/C) and 6707665(A/G) are negative, low quality or no-calls?

None of those SNPs appear in my results. I don't know whether that means no-calls or something else. I just downloaded my BAM fill and wll send it off to YFull later today.

So in the meantime my theory about the Boii is on indefinite hold!

Thread coverage is pretty good, so negative is assumed for all of them.

Richard and Petr,

I just checked again and S12993 is a no-call on my kit. I had previously consulted Astrid and Thomas Krahn about that SNP and received the following:

"S12993 is 91.6% similar to Chr2:133022457..133023469. I've designed primers, but the reverse primer isn't very strong. The run might fail. I wouldn't recommend the SNP if it isn't very important for you."

I emailed them again this morning to see if they thought there would be any value in ordering it. I'll let you know.

If your bam has you as 17760314=T, then you will likely also be S12993-.

My YFull results are still processing, but I can browse SNPs and am negative for all of those listed above. My placement on the tree above Petr is consistent.

most of this stuff is way over my head, but I was wondering if anyone knew a couple of key big Y snp's that a L513 kind of guy might have...I know how to check for that at least

well, according to MikeWWW newest L513 tree, me and the Vances have a new SNP FCG 16870!

I have no idea what that means, I have not had a chance to talk to any of the experts yet, and there is one more of my NPE Vance cousins that should get their Big Y results soon and thus let us triangulate our results fully

but I think this means we have a new SNP, perhaps a 'family' one, perhaps larger-they think we are an early break off from L513 so I really hope they can give us more info on that aspect

anyway, a few steps forward it seems
