We're still waiting on a paper to release with qualifiable samples for G25.

If you know of any Sudanese/Nubians willing to participate, feel free to PM me. I have a couple free FTDNA kits.

Thanks for the followup man! -- that's a big win

Feel free to share any findings, I'll shortly add them on the PCA with "deep-layer" runs.

Curious to see how their HG-like affinities will compare with other Bantu S.E./S.W. samples.
No problem at all man. I ran through them really quick last night and was surprised at:

Some of the Mozambicans harboring significant amounts of S.Sudanese 7+11/Experimental Basal East African/Mursi - this adds credence to a Great Lakes dispersal point from Zone J to Zone S I guess...seems that Nilotic admixture had already occurred to a degree before migration southwards? [on another note I still can't get my head around Cameroonians and their small Dinka-like ancestral component, is this something fixed in Bantu/Grassfields peoples or is it something more recent].

Pretty high levels of whatever Ken_LSA acts as a stand in for, higher than in the Cameroonians I believe.

One or two of the Angolans plotting very closely to the Esan/Yoruba with minimal HG drift and higher ANA than from what I've seen in other Bantu speakers [but not higher than in Yoruba/Esan or some of the NW Cameroonian individuals] with the experimental sims. Wonder if they can act as the closest thing to pristine Narrow Bantu speakers we have right now or would that still be some of the Grassfield's Cameroonians minus HG affinities.

edit: checked again and the Angolans are pretty mixed up with various HG streams as far as I can tell.

And how do they compare?
Yes, rs334 (sickle cell allele) most likely is from Africa. It is really a very bad marker of that, though, because it is under strong selection. When there is falciparum malaria around it is under strong positive selection up to a certain frequency (because having one copy is protective). When there is no falciparum malaria it is selected against (because having two copies gives you sickle cell anemia). When there's falciparum malaria around but you already have certain other alleles that protect against malaria and cause blood diseases, it's selected against, because they conflict with each other. So you could have the tiniest trace of African ancestry and end up with a high frequency of sickle cell allele, or start with lots of African ancestry but end up with zero sickle cell allele.

The case of the light skin variant rs1426654 (in SLC24A5) is similar in that it is under selection, so it's frequency won't reflect the original admixture proportion in the long run, though you wouldn't expect the selection to be nearly as intense as for sickle cell. E.g. in South Sudan, where people are very dark, there seems to be strong selection against light skin, so you'd expect it to fall in frequency there; whereas it seems to have increased significantly in frequency in Khoisan people from subtropical South Africa, where it is more common than you'd expect based on their proportion of West Eurasian-like ancestry.

Also, what is the actual relationship between the ancestral and derived variants of this rs strains? Where did the mutation of occur? Did it occur just once and then gets diffused to every person who has it now?
In principle the mutations could have occurred more than once, and multiple origins have been suggested for sickle cell allele, but last I heard both are thought to have a single origin. This is based on the haplotype background (pattern of other mutations around the mutation of interest).

By the way, in respect to the comment i made earlier on this thread, the absence or near absence of Anatolian and Iran neolithic like ancestries in cushites(except for some groups with known post neolithic Arabian admixtures) is highly significant for the biocultural history of NE African quadrant. Highly significant in deed. And I await preneolithic/neolithic DNA samples from Egypt, Libya and the Sudan to better understand this importance
I think Horners do have Anatolian_N ancestry via Levantine-like Farmers. The PPNB & PPNC samples are nearly half-Anatolia_N and I think the West Eurasian ancestors of Horners were similar with a lot less Anatolian Farmer ancestry possihly with similar amounts of ANF ancestry found in BedouinB & Mahra samples. When I replace Natufian with BedouinB or Mahra on G25 for Horners, the fits are still quite close.

The Zanji rebellion in Iraq took place in the 800s, which means slaves from the Zanj coast had already begun to reach the Gulf ~200 years before this TMRCA.

If the TMRCA was earlier, sat 2000 years ago, we could speculate about a link to ancient Azania or direct gene flow from the HOA.
@gihanga , yfull updated our tmrcas. 1800ybp for Arabs and 2700ybp for myself and them. I'm inclined to think, this wasn't the slave trade. Still doesn"t prove much, just my theory.

TmRCa between these Rwandan tutsi lineages is 3700ybp. Which sorta predates some of the PN samples. So an earlier split with PN groups from Kenya and Tanzania is looking more plausible. We will know for sure when yfull includes PN ydna samples in their tree. They told us to wait for our turn when asked...apparently there is a queue of paper/samples before us. Though the Viking samples got there super quick, makes me feel like a 2nd class citizen up these DNA streets

"We also investigated the Mota genome for a number of phenotypes of interest (6). As expected, Mota lacked any of the derived alleles found in Eurasian populations for eye and skin color, suggesting that he had brown eyes and dark skin. Mota lacked any of the currently known alleles that give lactose tolerance, which may have implications concerning when pastoralism appeared in southwestern Ethiopia. In addition, Mota did possess all three selected alleles that have been recently shown to play a role in the adaptation to altitude in contemporary highland Ethiopian populations (19). The presence of these mutations supports our conclusion that Mota is the descendant of highland dwellers, who have lived in this environment long enough to accumulate adaptations to the altitude "

Been looking at the PCAs to see if there would be anything closer to a 'pure' Bantu that could be used instead of Esan/Yoruba. On the G25 it doesn't make that much of a difference cuz that bimodal Atlantic vs Bantu mixture model doesn't really come across like it does on structure/admixture runs in most papers, except in very particular PCAs. But I found this individual who seems to be almost pristinely Bantu:

    Target: Cameroon_Mbo:CAM-MBO-019
    Distance: 2.3915% / 0.02391469

    2.2 Basal_East_African
    1.2 Ancient_Arabian
    0.8 Basal_Central-East_African
    0.4 Basal_South_African

The noise levels of HG ancestry are fascinating for a Cameroonian ethnic group. I wonder if Ekoid/Enjagham folk in South-East Nigeria would be quite similar to this profile. Unfortunately there isn't even a pristine Atlantic/Jola type individual ancient or otherwise in any data set let alone Bantu so the waiting game is killing me.

Started using the simulated samples of Natufians & ANA and it seems the ANA eats up all the mota-like affinities in Somalis while Mota ancestry remains constant but slightly reduced among the Borana and other Ethio ethnicities. One of the tech-savvy Africans on this forum should create a simulated AEA.

Yeah, I guess that's a possibility -- some degree of Nilotic-like admixture within the great-lakes prior to a further S.E. migration... you'd have to also take into account minor Nilotic-like ancestry which was already embedded in West-African Bantu pops before their mass-migration.
Lastly, the Nilotic-related ancestry within South-Cushitic speakers who would've hypothetically been absorbed along the way from Guthrie's "Zone J" (Great Lakes/Rift-Valley region).
It's worth adding, the Eurasian affinities related to absorbed pastoralist South-Cushitic speakers isn't apparent within these G25 scaled results -- though you can tease it out using unscaled cords

Depends which Cameroonians we're talking about, if we're talking far northern Cameroonians, there's plenty Central-Sudanic and Chadic-speakers who still harbour significant Ancestral East African (AEA) affinities from earlier periods. Actually, minor ~+- <5% AEA-affinities right across West-Africa, especially along the Sahel, is pretty standard. So, this shouldn't be a complete shocker in S.E/S.W. Bantu.

Yepp, this is to be expected from absorption of indigenous HG pops (KEN_LSA is a variant of East-African HGs).

Wonder if they can act as the closest thing to pristine Narrow Bantu speakers we have right now or would that still be some of the Grassfield's Cameroonians minus HG affinities.
Interesting you mention that: relating to the notion of a "pristine Bantu", I was curious to see the minor HG-related ancestry you're referring to -- there's quite a lot showing none or next to none:
