    Hi all,

I'm looking for some advice on Y-DNA testing. So far, I've been able to whittle down my haplogroup through selective SNP testing with Y-SEQ. It's been helpful. but essentially I still haven't found an answer to where my gr-gr-gr-gr grandfather was from (which is primarily what I'm looking to resolve). Would a big-Y test with a company such as FTDna help with something like this?

    Many thanks!

WGS 30X from Nabula Genomics or Dante Labs, they are cheap when on sale.

Sound advice if cost is a concern, it'll be well worth your patience if you can wait for the sale.

Many thanks both for the advice - much appreciated!

I see that Nebula have a sale on atm, so I may explore further.

Would you still recommend WGS even if, at this point, I'm just interested in greater resolution on my y-dna? The other option I was contemplating was something like FTdna Y-111 as I heard that I'm more likely to get indicative matches on that platform..

STRs only give you rough idea because of reverse mutation, SNPs are the King. Do the ones you want to compare with already have have 111 STRs test done?

A WGS test will get you yDNA, mtDNA and aDNA, also most but not all the 111 STRs. Big y-700 will give all 111 STRs and yDNA. YSEQ WGS400 test will give you yDNA, mtDNA, aDNA and all 111 STRs. But remember YSEQ WGS400 for $399 is only 15X, while Nabula Genomics WGS for $299 is 30X. Also note that WGS test is superior to Big Y test when it comes to SNPs. Hope it helps.

I would say go for Nebula since its WGS is the cheapest option but also not bad in terms of timing. FTDNA has a sale going on right now for Y-seq, u can check that out as well.

if you get Y-111 you wont get your subclade- to get your subclade is only with bigY. you will still have a basic haplogroup waiting - but you will get your matches best thing is if you want matches with your full new subclade is with Big-y700.

I take it you're trying to infer where your 4x great grandfather lived based on information from his closest matches.

If that's the case then as subpar as FTDNA's database is for South Asians, it's probably your best bet considering the strong priority you appear to place specifically on matching rather than the amount of data you get for your money (I apologise if my inference is wrong here).

With FTDNA's Big Y-700, you can currently get it for $379 as it's currently on offer for the next 4 days as of the writing of this post (offers from FTDNA occur throughout the year so you'll never have to wait too long for another offer to pop up). Unfortunately, you have to pay $99 to download the BAM file (absolutely asinine if you ask me) however you do get the Y111 service included in the price as well as YFull-esque interpretation of your Big Y-700 raw data (novel SNP discovery, haplotree placement, extraction of Y-STR's not included in the main Y111 panel) which is useful for comparing your STR's to other FTDNA customers (whether they show up as your matches or are on public FTDNA projects) who haven't purchased a Big Y.

Bear in mind that splitting payments by purchasing a Y111 initially followed by an upgrade to Big Y-700 is an option too if purchasing a Big Y-700 upfront (with no previous Y-STR panel purchases from FTDNA) is too cost-prohibitive, although the method I just mentioned will cost more in the long run so I recommend purchasing a Big Y-700 upfront if you can afford it. Generally, I don't recommend the Big Y-700 to South Asians as the main advantage they hold IMO is their matching database which isn't particularly useful for South Asians in any case however, since matching appears to be an exceptionally significant priority for you (every little helps, I guess), FTDNA could be a viable option specifically in your case.

Nebula Genomics constantly mentions that they have a partnership with FTDNA in the works however there's currently no ETA on this, and if/when the partnership comes to fruition, there will probably a sizable fee to pay for transfer to FTDNA on top of the $319 spent on Nebula (assuming you go for the 30x test) even though you get the advantage of being part of a matching database otherwise exclusive to FTDNA customers. They also have their sequencing performed by a company with ties to the Chinese government - some people are concerned by this for a variety of reasons so it's worth mentioning.

The other direct-to-consumer WGS vendors (i.e. Dante Labs, YSEQ, FGC) don't offer a pathway to FTDNA's database, however WGS tests have their own advantages (free mtDNA sequence plus the possibility of generating accurate superkits for GEDmatch/Admixture Studio/Global25 use) and you can still upload to YFull with them and match samples there. Many Big Y-700 customers upload their results to YFull as well as people who have purchased companies other than FTDNA.

If you have any questions (i.e. about terminology I've used) then I'm happy to clarify further.

Nebula's 30x WGS has a regular price of $288.99 including a 1 month membership (which can be cancelled before the second month begins) and the ENDOF2020 discount code which I would say is likely to be as low as it gets for the near future at least.
